南京地铁S6号线于2021年12月28日运营。

标志色为丁香色。

途经栖霞区、江宁区和镇江市句容市。

经过马群站、百水桥站、麒麟门站、东郊小镇站、古泉站、南京猿人洞站、汤山站、泉都大街站、黄梅站、童世界站、华阳站、崇明站、句容站。

全长43.7公里，其中地下段16.26公里、地面段1.36公里、高架段26.08公里。

共设13座车站，其中地下站7座、高架站6座 。

线路采用4节编组B型列车。

最高运行时速120公里/时。
